Description

Dimethocaine, commonly known as DMC or lidocaine, is a chemical having a stimulatory action. This impact parallels that of cocaine, however, dimethicone seems to be less strong. Just like cocaine, dimethicone methane is addictive owing to its activation of the reward circuit in the brain.

However, dimethicone is a legal cocaine replacement in some countries and is even listed by the European Monitoring Centre for Drugs and Drug Addiction (EMCDDA) under the category "synthetic cocaine derivatives". The structure of dimethicone, being a 4-aminobenzoic acid ester, resembles that of procaine. It is found as a white powder at room temperature.

When a product marketed online in the UK in June 2010, labeled as dimethicone was analyzed, it was revealed to be a blend of caffeine and lidocaine, and the absence of any dopaminergic stimulant element in such mixtures may explain the limited recreational effects claimed by many consumers.

Other samples tested have nevertheless been proven to include real dimethicone, and one branded "bath salt" product containing predominantly dimethicone as the active component, was observed to have been especially vulnerable to misuse by intravenous drug users in Ireland.

Methacryloyl chloride (6.0 mmol) was added dropwise to a mixture of dimethocaine (5.0 mmol), NEt3 (1.4 mL, 10 mmol) and DMAP (30 mg, 250 μmol) in CHCl3 (10 mL) at 0°C. The reaction mixture was heated to room temperature and stirred until the starting material was completely consumed (1-3 hours). H2O (about 20 mL) was then added to the reaction mixture and the reaction mixture was extracted with CHCl3 (about 20 mL x 2). The organic fractions were combined and dried over Na2SO4. The organic fractions were concentrated in vacuo and a solution of the resulting residue in dry THF (10 mmol) was added dropwise to a suspension of NaH (60% dispersion in paraffin, 240 mg, 6.0 mmol) in dry THF (10 mL) at 0°C. The resulting mixture was stirred at room temperature for 5 minutes, and then halogenated aromatic hydrocarbon (0.7 mL, 5.5 mmol) was added dropwise to the solution at 0°C, and the solution was stirred at room temperature overnight. An aqueous solution of NaHCO3 (20 mL) was added to the reaction mixture. The mixture was extracted with EtOAc (30 mL × 3), the combined organic extracts were dried over Na2SO4, and the combined organic extracts were concentrated in vacuo. The residue was purified by silica gel column chromatography using a gradient of n-hexane/EtOAc = 12-100% to obtain the target product molecule. [2]
