Clomiphene citrate, commonly known by its brand name Clomiphene, is a selective estrogen receptor modulator (SERM). It is used primarily in reproductive medicine to stimulate ovulation in women who have difficulty conceiving due to anovulation or irregular ovulation.

Clomiphene is a nonsteroidal triphenylethylene derivative that acts as both an estrogen agonist and an antagonist, a selective estrogen receptor modulator. Its estrogen agonist effect is only manifested when estrogen levels in the body are extremely low. At normal estrogen levels, clomiphene mainly acts as an anti-estrogen. It is usually taken orally once a day, and the course of treatment usually lasts for five days.

Medical Use

Clomiphene is a drug used to induce ovulation, and is indicated for infertility due to anovulation or oligoovulation. There is insufficient evidence for the efficacy of clomiphene in female infertility without a clear cause. Some studies have found that the clinical pregnancy rate with clomiphene is 5.6% per cycle, while the pregnancy rate without treatment is 1.3%–4.2%.

The timing of the drug is important: it is usually recommended to start taking it on the fifth day of the menstrual cycle and to maintain regular sexual intercourse.

Ovulation induction: Clomiphene is mainly used to induce ovulation, especially for women with infertility due to anovulation or oligoovulation. It stimulates the development of follicles in the ovaries and ovulation by inhibiting estrogen receptors in the hypothalamus, thereby promoting the release of follicle-stimulating hormone (FSH) and luteinizing hormone (LH) from the pituitary gland.
